Position title: Scientist I/ Associate Scientist IIDepartment name: Process Cell CultureLocation: Gaithersburg MDPosition ID number: 3510-276Position description:You will develop stable production cell lines for therapeutic antibodies or other protein pharmaceuticals using mammalian cell lines (CHO and NS0). You will be a leader in technology development projects including microarray profiling of cell lines to diagnose expression bottlenecks developing high-throughput robotic procedures for clonal cell line isolation expansion and evaluation flow sorting to enrich high-expressing populations and developing automated data management systems. You will maintain knowledge of current cell culture literature and present and publish your results inside and outside MedImmune. Your responsibilities will include adaptation of cells to animal-component-free media sub-cloning cells and selection of the highest expressing clones. You will expand the cultures from the single cell stage in semi-solid media up through 96-well plates into suspension in shake flasks. You will conduct production model experiments by feeding the best cultures over 2-3 weeks. You will determine cell number and protein production levels and will test the best clones for their ability to express high protein titers for up to 100 population doublings. You will prepare cryopreserved banks of the best clones and test various biochemical properties of the proteins expressed. As a team player in our department you will maintain the laboratory and its equipment provide technical support for upstream processes in therapeutic protein manufacturing and pack and ship cell samples to outside vendors for testing. You will keep detailed and accurate records of your work. You will generate and review SOPs and batch records and assist in other areas as needed including operating bioreactors. Position requirements:Extensive experience in cell and molecular biology including mammalian cell culture and stable cell line generation. Experience with robotics high-throughput processes and automated data management. Experience with microscopy ELISA flow cytometry. High level of scientific curiosity and excellent problem-solving organizational and documentation skills with a strong focus on details and results. Quantitative technical writing and oral presentations skills are essential. Proficiency in MS Office computer programs including word processing spreadsheets and databases required. Must be able to work for multiple hours in the lab. The job involves pipetting and other repetitious operations. Periodic weekend work is required.Educational requirements:For Scientist I: Ph.D. in cell biology microbiology biochemistry molecular biology or biochemical engineering or related disciplines for 0-3 years or M.S. degree in one of the above areas plus 8-10 years industry experience or B.S. degree in one of the above areas plus 10-13 years industry experience.
