Basic histology for a light microscopy: This includes a number of techniques for research and clinical testing, (includes cutting controls) including but not limited to immunofluorescence, immunoperoxidase, laser capture micro-dissection (LCM). Techniques may include standard and non-standard protocols as needed for a variety of routine and non-routine, including tissue processing, microtomy, staining, including RNase free and DNA free histology for various applications. Processing/sectioning is specific to needs of specimen, final need of the project or researcher. Perform immunohistochemistry procedures by programming (entering patient data) automated IHC stainers, via computer for daily staining runs. Label immunohistochemistry slides, patient and controls, for daily runs. Extreme accuracy and concentration required. Dry, deparaffinize, hydrate and pretreat (antigen retrieve) slides as required by protocols. Set up IHC stainers with appropriate vials of antibodies, reagents, solutions, and make dilutions as needed. Load IHC stainers with slides/reagents in specific order. Remove from stainer following completion of staining run, dehydrate, clear, coverslip. Perform special stains as needed. This may be manual or automated. Make up needed reagents for special stains. Clean all equipment used (special washing). Adhere to all safety, biohazard regulations. Proper disposal of all reagents, chemicals and solutions via regulations of the University of Louisville DEHS. Review controls and specimens for proper staining. Report to supervisor and pathologist slides not properly stained and set up for restaining. Problem solve any noted lack of conformity. Note detail for why procedure not working Snap Freeze, cut, stain muscle biopsies, using tedious enzyme histochemistry protocols. Snap Freeze, cut, stain sections for other applications including immunofluorescence such as skin, and minute kidney biopsies and LCM. Give factual information on phone about lab procedures, tissue preparation to associates, clients and students. Communication with researchers over projects. Implement new procedures as formulated by supervisor. Maintain established database for all specimens received into the labs, and discharged from the labs. Back up lab clerk phone coverage, logging in specimens, making slides.
